細胞融合をもちいた軸索輸送の分子機構の研究

研究概要	軸索輸送のメカニズムはかなり調べられてきたが、その生理的機能はまだ解明されていない。この研究では軸索輸送の生理学的な面の解明に主点をおいた。神経細胞では、神経化学物質の生理的作用が一番問題となるそこでまづ神経化学伝達物質がどのように軸索輸送に影響をおよぼすかをみた。マウスの上頚神経節の培養細胞の軸索輸送をビデオ増感顕微鏡像で観察すると多くのオルガネラが上行性或いは下行性に0.5μ〜1.0μm/秒で輸送されているのがわかる。この細胞に1μmのアセチルコリンを投与すると運ばれている粒子数が序々に減少していき数分以内に輸送が停止する。この停止の状態は濃度に依存する。そしてアセチルコリンを除去するとすぐに軸索輸送には復帰する。この伝達物質の効果は可逆的であった。また同様な効果が脊髄後根神経節細胞でもみられた。後根神経細胞にGABAを投与すると可逆的に軸索輸送が停止するのが観察された。つぎにアセチルコリンがどの様に軸索輸送に作用するかをみた。まずアセチルコリンリセプタ-にムスカリン性に効くのかニコチン性に効くのか調べた。ムスカリン性リセプタ-のアンタゴニストであるQNXを作用させると軸索輸送停止の効果が消失する。そこでこの輸送停止効果はムスカリン性であると推論できる。しかしQNXは量が多くなるとニコチン性リセプタ-にも効果があるのでこの点現在研究中である。次にPIP_2に作用してDGとIP_3によるPLCを抑えるNCDCの作用を調べた。NCDCはアセチルコリンの軸索輸送停止効果を抑える作用がある。このような実験事実からアセチルコリンの軸索輸送に対する作用はイノシト-ルリン脂質系が関与することが考えられる。そうするとCa^+の軸索輸送に対する作用が問題となってくる。この点についてFura2を用いて現在研究を進めている。
