研究概要	リソゾ-ムシステインプロテア-ゼ群は他のリソゾ-ムの可溶性酵素と同様、ゴルジ装置で分泌蛋白、細胞膜蛋白と仕分けされ、その後リソゾ-ムへ包み込まれることがこれまでの研究でわかってきた。最近カテプシンHとLのcDNAクロ-ニングと塩基配列の決定、遺伝子構造の決定を行い、3種のカテプシン(B、H、L)のプレプロ部分を含む全一時構造が判明した。その結果細胞内局在化にマンノ-スー6ーリン酸と共にプロ部分が仕分けの信号かどうかをしらべる手がかりを得た。今年度は1)0ーフェナンスロリン処理によりプロ酵素から成熟一本鎖酵素への変換が抑制されるが、カテプシンDの阻害剤であるペプスタチンによっても抑制されることを確認した。プロ酵素から一本鎖成熟酵素への変換を非常に短いパルス実験で行うと中間体と思われるものが少なくとも2バンド認められたので、このプロセスは多段階である可能性が示唆された。IーCell病患者の線維芽細胞やEー64処理細胞でも中間体が検出でき、プロ酵素から成熟酵素への変換には数種類の酵素(金属およびアスパルテックプロテア-ゼ)が関与すると想定される。3)0ーフェナンスロリン処理細胞ではゴルジ装置にプロ酵素のまま蓄積していることをパコ-ル密度勾配遠心および免疫電顕で確認した。4)cDNAより得られた結果をもとに、合成ペプチドを作成し、プロカテプシンBおよびLにそれぞれ特異的な抗体を得た。いずれも成熟酵素とは全く反応しない。プロカテプシンBおよびカテプシンBの抗体を用いて甲状線におけるそれらの局在を免疫電顕(金コロイド法)で調べたところ、カテプシンBはリソゾ-ムに多数のゴ-ルド粒子の集積がみられたが、プロカテプシンBがゴルジ装置と思われるところに弱いゴ-ルドの集積が観察できた。従って変換の場はゴルジ装置からリソゾ-ムまでの過程であることが確認された。プレリソゾ-ムではいづれの型であるか現在追求中である。
