正常組織におけるPAFとその内在性インヒビタ-

研究概要

PAF(血小板活性化因子)は免疫・炎症反応のメジィエ-タ-と見なされ、一般には正常組織には存在しないと考えられていた。然しながら、我々は外界から何らの刺激を加えない正常組織例えばラット子宮にも微量ながらPAFが存在することを初めて報告したが、その生理的意義については全く不明である。今年も引き続き正常組織のPAFについて、また他方、疾病とPAFとの関係について以下の実績を得た。
1.ラットおよびモルモット心筋に存在するPAF類縁化合物について
いづれにもPAF、アシル型PAFおよびビニ-ル型PAFが存在し、このビニ-ル型が発見されたのはこれが始めてである。量的にはアシル型が一番多く、次いでビニ-ル型そしてPAFの順であり、いづれも16:0が主成分であった。
2.ラット胃におけるPAF類似化合物について
胃では胃前庭、体部、前胃の順に、十二指腸では胃体部より多量にPAFおよびアシル型PAFが存在しており、アルコ-ルの経口投与によって著明に低下した。この際アセチ-ル・ヒドロラ-ゼ活性は上昇していた。正常組織におけるPAFとその関連化合物の意義は不明ではあるが、オ-タコイド様の機能を持っていると考えている。
3.アレルギ-と関係の深い鼻ポリ-プの中に、好酸球および好中球の含まれた胸水の中に、PAFおよびアシル型PAFを検出し同定した。また人単核球は殆ど特異的に尿酸刺激にてPAFを生成した。
以上の様にPAFは炎症免疫反応に関係するのみならず、正常組織の機能にも深く係わる重要な生理活性物質である。
