T細胞の自己認識機構と抗原レセプタ-:T3複合体遺伝子及び分子構造の比較研究

研究概要

1.胸腺細胞におけるCD3分子を介するシグナル伝達系の発達が、特定のマウス系統の胸腺で円滑に進まないことが判明した。この発見は胸腺細胞の機能的分化もT細胞レパ-トリ-の選択と同様胸腺基質細胞に依存することを世界で初めて明かにしたものである。
2.胸腺細胞の分化に伴なう表現型の変化を詳細に解析し、これらの過程が従来考えられてきたより著しく複雑であることを明らかにした。
3.胸腺細胞の自己寛容の成立過程を解析し、自己抗原反応性T細胞が胸腺内で消去されるメカニズムを明かにした。この消去には、骨髄由来Ia陽性細胞が重要で、消去は胸腺髄質内で胸腺細胞分化の限られた時期に起る。さらに胸腺上皮細胞は自己寛容誘導には直接関与していなかった。
4.骨髄移植後に生ずるGVHRによって自己寛容が破綻することを発見した。これにより、従来GVHRの末期に認められるランド病が自己免疫によることが示唆された。
5.特定のH-2クラスI分子によって、T細胞のポジティブセレクションが生ずることを明かにした。この選択に関与する分子をH-2ミュタントマウスを用いて解析した結果、クラスI分子のα1鎖に機能部位のあることが判明した。
6.合成ペプチド抗原に対するT細胞反応をH-2リコンビナントマウスを用いて解析した。その結果、T細胞の抗原識別に関与するH-2クラスII領域の抗原結合部位が判明した。又、異なるクラスII分子間においても、その機能部位が比較的良く保存されていることが明らかとなった。
