条虫感染における宿主-寄生虫相互関係の免疫学的・分子生物学的研究

研究概要

平成元年度は各種条虫卵の免疫原として重要性ならびにワクチン抗原としての可能性について実験モデルを用いた基礎研究を試み、以下の成果を得た。
1.マウス小腸組織内へ侵入したH.microstoma六鉤幼虫の時間的・空間的挙動について:a)虫卵投与後15分以内で小腸組織内へ六鉤幼虫は侵入する。b)六鉤幼虫は侵入数は虫卵投与後1時間で最高となり、2時間で小腸組織内から姿を消す。c)肝臓組織内には六鉤幼虫は侵入していない。d)六鉤幼虫の侵入しやすい部位が小腸組織に存在することが、蛍光抗体法などにより明らかになった(蛍光抗体法は落射蛍光顕微鏡による)。
2.H.diminuta六鉤幼虫の小腸組織内への侵入:63年度の成果の一つとして、中間宿主と終宿主を異にするH.microstoma六鉤幼虫がマウス小腸組織内へ侵入する事を世界で初めて明らかにしたが、H.diminutaにおいても多数の六鉤幼虫がマウス小腸組織内へ侵入しているのが確認され、これらの現象は普遍的であることが明らかになり、条虫-宿主間の免疫応答と適応戦略の機構解析に極めて有効な系であることが明らかになった。
3.H.nana近縁種H.microstoma、H.diminuta、T.taeniaeformis虫卵投与マウスが、H.nanaの攻撃感染に示す抵抗性:H.nana虫卵攻撃感染に対する非常に強い再感染抵抗性は同種六鉤幼虫によってのみ惹起されるが近縁種六鉤幼虫の小腸組織内侵入によってもかなり強い抵抗性が惹起されることが明らかになった。
4.条虫分子ワクチンの作製:T.taeniaeformis六鉤幼虫mRNAをもとにしてcDNAを作製し、そのEcoRI切断断片をプラスミドに組み込み発現させることにより、F1〜5のクロ-ンを得ることに成功した。その中でF3ワクチン1μg接種マウスがT.taeniaeformis虫卵攻撃感染に対し98%の強い発育阻害を示した。
