研究概要

細菌学的、免疫学的な方法を用いて様々な歯周疾患の病態解析を試み、多くの成果を得た。その概要は次の通りである。
1)7種類の歯周病原性細菌に対する重度歯周炎患者の血清IgG抗体価をELISA法により測定し、歯周疾患の進行度との関連性を検討したところ、B.gingivalisとA.actinomycetemcomitansが重要な役割を示していることが示された。2)歯周組織破壊における白血球機能の関与を調べる目的で、ダウン症患者の末梢血中の多形核白血球の走化能を測定したところ、多形核白血球の機能障害がダウン症患者の歯周組織破壊に関与していることが示された。3)活性化脂質である血小板活性化因子(PAF)について調べたところ、ヒト炎症性歯肉中に検出され、歯周疾患における炎症反応にPAFが関係することが明確にされた。4)末梢血多形核白血球機能に関連してアラキドン酸代謝における15-HETEの産生能を定量したところ、若年性歯周炎及び重度進行性歯周炎患者では産生能低下がみられたことから、これらの患者の末梢血多形核白血球の機能低下と関係が深いと考えられた。5)各種歯周病原性細菌の抗原性をImmunoblot法を用いて分析した。B.gingivalis W50、Veionella parvula ATCC17745、A.actinomycetemcomitans ATCC9710については全菌体とouter membrane proteinsに分けて分析し、B.gingivalis 381株については全菌体を用いて分析した。この結果から、多種類の抗原蛋白質に対して患者血清の強い反応が認められ、強い抗原抗体反応を示した各種細菌の抗原蛋白質に関する検索の重要性が示唆された。6)A.actinomycetemcomitansの抗原物質を明確にする目的でA.a.菌Y4株の抗原遺伝子のクロ-ニングを行い、抗原を発現している遺伝子として5つのクロ-ンを得ることができた。そしてクロ-ニングによる分析を行なうことは今後、DNAプロ-ブやワクチン作成に寄与すると考えられた。
