研究概要	1)精製したマウスHSP90を未変性ゲル電気泳動したところ、2本のダイマ-と1本のモノマ-に分れた。それぞれのバンドを切り出し、6%SDSーPAGEで調べたところ、α/αおよびβ/βのホモダイマ-とβモノマ-であることがわかった。細胞抽出液を未変性ゲル電気泳動し、抗HSP90抗体でウエスタンブロットすると、やはり2本のダイマ-と1本のモノマ-が検出された。 2)比較的低イオン強度の緩衝液で抽出した細胞抽出物をショ糖密度勾配遠心し、特異的基質ペプチドを用いてカゼインキナ-ゼ(CKII)活性のサイズを調べた。CKIIのピ-クは約8Sで、抗HSP90抗体を加えると10Sにシフトした。この結果は8SーGRと同じように、CKIIもHSP90と複合体を形成していることを示唆する。さらに、精製したCKIIを但イオン強度溶液中に保つと会合体を作るが、HSP90を添加すると会合体は解離し、CKII活性は8Sになる。このとき、可溶化にともないCKII活性が著しく上昇することを認めた。 3)熱ショックによって、おそらくアクチンを核内へ運ぶタンパク質であるコフイリンの分子的解析を行った。N端より30番目からはじまるKKRKK配列をKTLKKに改変し、マウスC3Hー2K細胞に発現させた。N端に9個のアミノ酸からなるペプチドが余計に付くように工夫がしてあり、そのペプチドに対する抗体で変異コフイリンを内在性コフイリンと区別して局在を調べた。KKRKKコフイリンが熱ショックによって核内移行するのに対し、KTLKKコフイリンは核内移行しなかった。KTLKKコフイリンは細胞質ロッドを形成するので、KTLKKは核内移行シグナルであると結論できる。
