コムギ・エギロプスの葉緑体ケツムにみられる変異集中領域の構造解析

研究概要

当初予定していたコムギ・エギロプス属植物の5種(Triticum aestivum,Aegilops crassa,Ac.sequarrosa,Ac,candata,Ac.ovata)の変異集中領域約4.5kbpのDNA sequenceを決定し、DNA構造を詳細に解析した。タバコの相当する領域と比較することにより、コムギ固有な遺伝子再配列を発見した。これは遺伝子の転位であるが塩基配列の比較からこの転位は短い(4ー12 nuculeotides)反復配列を介した非正統的組み換えによるものであると結論づけた。また、コムギ・エギロプス属の葉緑体DNA相互、コムギグル-プとイネの塩基配列を比較して、それぞれのゲノムでみられる欠失には、短い反復配列(5ー20bp)を介したものも含まれ、DNA複製酵素・DNA組み換え酵素のそれぞれが欠失に関与しているものと結論づけた。葉緑体ゲノムでみられる欠失の機構は、バクテリア・プラスミドのそれと対応していた。この領域全体はATーrichであり、哺乳動物ゲノム中に散在してみられるSINES(Short Interspersed Repeated Sequences)配列と構造上の共通点がみられた。これらの葉緑体ゲノムの変異集中領域と相同な配列がミトコンドリア、核ゲノムでも再編成した形で検出された。このことは、この領域がゲノムをこえて転位していることを示唆する。この領域には、偽rpl23遺伝子も含めて5つの遺伝子が存在する。各遺伝子の塩基配列を比較することにより、コムギ・エギロプス属という近縁な植物の葉緑体DNAに働く選択圧を特徴づけた。さらに、この領域のそれぞれの遺伝子に異なる陶汰圧がかかっていることを明らかにした。DNA塩基の置換率から、コムギ・エギロプス属植物の分化した時期、コムギグル-プが他のイネ科植物のグル-プ(イネ、トウモロコシ、オオムギ)から分化した時期を推定した。
