無髄一次求心線維の脊髄内終末の研究

研究概要

本研究の本年度における研究目的は、痛覚の伝達機構を解明する上で必要な無髄性求心線維の解析、とりわけ、体性神経系と内臓神経系におけるシナプス機構の形態学的解析及びその比較検討であった。内臓性一次求心線維は脊髄内終止の分布において体性一次求心線維と違いがあることは既に昨年次報告してある。これを定量的に比較検討するために各々の線維、二例づつにおいて終末膨大の数を数えた。体性一次求心線維は1-2箇所の密な終末分布を示し終末膨大の総数は二例とも約1500であった。個々の神経線維の感覚受容野を同定していないので感覚特性については解らないが、比較的狭い範囲の二次神経細胞に対して強い影響を与えると考えられる。内臓性一次求心線維は終末膨大の総数は約6100と約5400個であった。しかし終末している分技の数が22本と18本と異なり、一分技当りの数では約280と300と近い値をとる。これは内臓性一次求心線維が広い範囲の二次神経細胞に対して比較的均等な影響を与えていると考えられる。この伝達様式の違いは腫瘍遺伝子発現蛋白であるC-fasの細胞免疫染色による所見と一致している。
標識した体性無髄求心線維終末の電子顕微鏡による観察では、脊髄後角のI-II層に存在する終末膨大はCentral型の終末を示していた。このシナプスは二次ニュウロンの樹状突起またはその棘突起に前シナプス結合をしており、又一方では起源は未定であるが介在ニュウロンと思われる円形小胞をもつ終末の後シナプス要素となっていた。体性無髄神経として標識された終末の中には有芯小胞とみられる顆粒はほとんど含まれていなかった。その結果おそらくこの例では一次求心線維終末はDense sinuso id axon terminal型の終末に分類されると考えられる。ただ現時点ではこの終末がどの様な伝達物質を持つかは色々の実験を試みたが未だ明らかにし得なかった。
