人及びラットの肝細胞癌,肝肉胆管癌の前駆病変細胞核DNA ploidyに関する研究

研究概要

先にNーαーfluorenylacetamideCFAA)の投与により誘発されるラット肝細胞性変異増殖巣細胞核DNA ploidyパタ-ンが非病変肝細胞核DNA ploidyパタ-ンに比し,拡がりを有し,DNA量の相対的増加を示すことを報告したが,その後の幾つかの報告は逆に変異増殖巣肝細胞がploidy reductionを有し,diploidの細胞が増加することを指摘している。我々は変異増殖単細胞核DNA量は病変の誘発方法により多様性を示すものと推論している。今回,2つの肝発癌モデルを用いて病変を誘発し,変異増殖巣細胞核DNA量の比較解析を行った。第I群では病変はSoltーFooberのモデルにより得られた(diethylnitrosamine 200mg/kg1回投与,0.02%FAA飼料投与,2/3部分肝切除を行った)。第II群では変異増殖巣は0.02%FAAの飼料投与のみになって得られた。この様なラットモデルにより誘発された肝病変を特に経時的に動物を屠殺した材料についてFeulgen染色切片標本にてmicrospectrophotomater(MMSP)を用いて比較観察した。第I群に出現した変異増殖巣はほとんどが好酸性型で出現頻度は経時的に減少し,diethylnitrosamine投与後6週から8週にかけて35.0/cm^2→15.8/cm^2の如く変化した。第II群は於いても出現増殖巣の多くは好酸性型であり、経時的な出現頻度の変化は第I群と異なり、FAA投与12週まで増加し、その後漸減するものであった。第I群に於いて,好酸性変異増殖巣は時間の経過と共に相対的DNA量の低下を示し,diploid化を惹起した。一方,第II群に誘発された好酸性変異増殖巣のDNA量の経時的変化は第I群と少々異なり,FAA投与6週後と16週では本質的な差異はみられなかった。第II群に主として観察されたtoluidine blue陽性の超塩基型変異増殖巣細胞核DNA ploidyパタ-ンはaneceploidyを示した。変異増殖巣の細胞核DNA ploidyパタ-ンは病変の種類の他,誘発モデルの相違,時間的経過等により異なりを示すことが確認された。
