癌患者単球・マクロファ-ジの抗がん活性発現能とその強化

研究概要	末梢血単球及び肺胞マクロファ-ジの抗腫瘍性並びにモノカイン(IL-1,TNF)産生能の解析を健常者および癌患者を対象に行い、次の点を明らかにした。1)健常人末梢血単球は活性化刺激にて抗腫瘍性を発揮するが、その機構の一つとして細胞由来のインタ-ロイキン1αが重要な役割を果たし、ガンマインタ-フェロンは一次刺激として作用し、抗腫瘍増強作用を示した。2)細胞結合型IL-1αは末梢血単球より肺胞マクロファ-ジ及びGM-CSFにて成熟した単球由来マクロファ-ジにより有意に多く産生された。3)肺胞マクロファ-ジ及び造血因子にて成熟した単球由来マクロファ-ジはIL-2活性化キラ-細胞により細胞障害を受けることを明らかにした。4)活性化単球はIL-1及びTNFとは異なる新規の抗腫瘍性モノカインを産生し、分子量30Kd、pI6であることを確かめた。5)癌患者末梢血単球は細菌内毒素刺激にて抗腫瘍性を発現するが、健常人よりその程度は低下しており、IL-1産生能も有意に低かった。しかし、多重層リポソ-ム内に封入したアジュバントにより活性化された癌患者単球は健常人のそれと同程度の抗腫瘍性を示した。6)肺胞マクロファ-ジは末梢血単球より高いTNF産生能を示すこと。癌患者と健常者群との間に肺胞マクロファ-ジによるTNF産生能には有意差がないことを明らかにした。以上の成績から、ヒト単球-マクロファ-ジは成熟度により異なったモノカイン産生能を有し、局所での生体防御において重要な役割を果している可能性が示唆された。
