^<10>Bーモノクロ-ナル抗体によるヒト黒色腫細胞の熱中性子捕捉効果の解析

研究概要	メラニン生成活性に依存することなく^<10>B元素を黒色腫細胞に集積させる目的で、黒色腫細胞関連抗原に対するモノクロ-ナル抗体(MoAb)に^<10>Bを結合させ、熱中性子照射で癌細胞を選択的に殺傷する研究を展開してきた。癌細胞1個当り2×6^<10>個分子の抗体が結合する。熱中性子捕捉療法成功のために必要とされる1細胞当り10^9個の^<10>B元素と結合させるに必要な1抗体当りの^<10>B元素数は10^3個である。本研究ではヒト黒色腫細胞に対するモノクロ-ナル抗体763、74Tとヒト黒色腫細胞P22を用いて、以下の結果を得た。 <方法と成果> 1.細胞当りの^<10>B元素数:P22細胞にビオチン化763、74T、更に^<125>Iラベルアビディンを反応させ、放射活性より1細胞当りのアビディン数を求めると3.84×10^6個となり、細胞当りの^<10>B数では1.6×10^8個となった。まだ目標とする細胞当り10^9個には不足である。2.熱中性子照射による致死:^<10>Bーアビディンとビオチン化抗体を反応させた後、P22細胞をテフロンチュ-ブに入れ、異なる熱中性子量を照射し、コロニ-法で生存曲線を得た。^<10>B処理をしていない抗体を用いた対照に比べ、Do値は小さく明らかな致死増大を得た。 <反省と今後の展望> 前年度に得た細胞増殖曲線の結果を支持する結果であった。しかし、^<125>Iを用いた分析では1細胞当りの^<10>Bが1.6〜10^8個とまだ少ない。従って今後は新たな抗体を見い出すこと、及び^<10>Bの定量を間接法でなく、より直接手法であるICPーMS法で行い、細胞当りの^<10>B元素数を求める。次にヒト黒色腫担癌ヌ-ドマウスに^<10>Bーアビディンービオチン抗体を投与し、腫瘍に集積する^<10>B元素量を実測し、さらに熱中性子照射で、腫瘍退縮効果を求めたい。
