顆粒膜細胞の増殖と文化におけるIL-1の意義に関する基礎的研究

研究概要

ブタ顆粒膜細胞の培養系を用いて、顆粒膜細胞に対するインタ-ロイキン1(IL-1)の作用について検討した結果,以下の知見が得られた。
1.小卵胞由来の未成熟な顆粒膜細胞のFSHによる分化に対してIL-1は抑制作用を示した。すなわちIL-1はin vitroにおいて,FSHによるエストロゲン合成酵素(アロマタ-ゼ)の誘導を抑制し,エストロゲン産生を減少させるとともに、プロゲステロン産生をも減少させた。またIL-1は,FSHによるLHレセプタ-の誘導に対しても抑制作用を示した。次に,FSHにより誘導されるステロイド産生に対するIL-1の抑制効果のメカニズムについて検討を加えた。その結果,まずIL-1はFSHにより誘導されるcAMPの産生を抑制した。cAMPの産生以後ステロイドの産生に至るいわゆる'post-cAMP steps'については,IL-1はエストロゲン産生(アロマタ-ゼ活性)に対しては抑制作用を示したのに対し、プロゲステロン産生に対しては影響をおよぼさなかった。
2.未成熟な顆粒膜細胞の増殖に対するIL-1の作用を、[^3H]thymidineの取りこみにより検討した。その結果,IL-1は濃度依存的に[^3H]thymidineの取りこみを増加させた。またインスリンやepidermal growth factor(EGF)といった,顆粒膜細胞に対する既知の増殖促進因子とIL-1を同時に添加したところ,IL-1はインスリンやEGFの増殖促進作用に対して相乗作用を示すことがあきらかとなった。
