研究概要	心脈管系は神経伝達物質やホルモンなどの化学受容と応答の場であるとともに、血圧や、血管・心臓の収縮力などの物理量の発生とその受容応答の場である。著者は、イヌの脳血管に機械的伸展刺激を与えて生じる(stretch activation)筋原性収縮の受容・応答機構について研究し、以下の結果を得た。1)細胞内Caシグナルと筋原性収縮の同時測定、蛍光分光測定法を用い伸展刺激の受容とセロトニンやエンドセリンなどの化学受容や膜の脱分極による収縮に関わるCaの動態の特性を明らかにした。即ち、伸展刺激による筋原性の収縮発生に先行して細胞内Caシグナルの上昇があり、また筋原性収縮の維持相ではCaシグナルの減少がみとめられ、収縮の発生初期相と維持相ではCaに対する収縮タンパクの感受性が変化する可能性を示唆した。このような細胞内Caシグナルと収縮に分離が観察されることはエンドセリン自身あるいはエンドセリンとセロトニンの組合せでも認められ、収縮系をはじめとする細胞内情報伝達系においてフォスファチジルイノシト-ル代謝を介したプロテインキナ-ゼC等が化学受容のみならず物理受容にも関係していると考えられる。この点に付いて今後さらに追求したい。2)筋原性収縮反応と内皮機能との関連、筋原性収縮反応は内皮の有無に拘らず、誘起される。このはイヌのみなず他の動物、例えばネコでも観察され、種を越えた脳血管特有な反応であることを改めて確認した。一方、酸化ヘモグロビンによる筋原性収縮反応の増強作用が内皮から遊離される液性因子によることをカスケ-ド法を用いた実験から証明した。この液性因子はトロンボキサン、プロスタグランジンF_2αとは異なることを薬理学的および放射免疫測定法を用いて調べた。以上の結果を国内外の学会に報告し、論文に発表した。
