DNA組換え乳酸菌を用いた齲蝕ワクチンの開発

研究概要	前年度において、我々はう蝕原性Streptococcus mutans MT8148株のタンパク質抗原をコ-ドする遺伝子(pac)のクロ-ニングに成功し、その全塩基配列を決定した。本年度は、このクロ-ニングしたpac遺伝子を乳酸菌の1種であるStreptococcus lactisに組み込んだ。すなわち、pac遺伝子のBamHIーSphI断片をレンサ球菌ー大腸菌間のシャトルベクタ-であるpSA3に連結し、キメラプラスミド(pSM1)を作製した。ついで、pSM1プラスミドのDNAでS.lactis IL1403株をプロトプラスト法により形質転換させた。その結果、約20個の組換えS.lactis株が分離された。これらの菌株がpSM1プラスミドをもっていることは、アイソト-プでラベルしたpac遺伝子のBamHIーSphI断片を用いたサザンブロット分析で確認した。ついで、組換えS.lactisがS.mutansのタンパク質抗原を発現しているか否かを、ウエスタンブロット法で調べたところ、これらの乳酸菌が同抗原を産生していることが確認された。同抗原の産生量をイムノドット法で調べたところ、S.mutansの産生量の約1/10程度であった。組換えS.lactisにおけるpac遺伝子の転写レベルをRNAドットブロット法で調べたところ、S.mutansの1/4〜1/8程度であった。S.lactis IL1403株中においては、pSM1プラスミドは比較的安定であった。以上のように、本年度はS.mutansのタンパク質抗原を発現する組換え乳酸菌の作製に成功した。今後、この組換え乳酸菌のう蝕ワクチンとしての有効性について検討する予定である。
