DNA組換え乳酸菌を用いた齲蝕ワクチンの開発

研究概要	前年度に、Streptococcus mutans MT8148株のpac遺伝子をシャトルベクーpSA3に連結した組換えプラスミドpSMlで、乳酸菌の1種であるStreptococcus lactis IL1403株を形質転換して、菌体内にPAcを産生する組換え株を作製した。本年度は、この組換え乳酸菌をホルマリンで殺菌したものを、BALB/cマウスに合計7回経口投与した。最終の経口投与から1週間後に唾液と血清を採取した。唾液および血清中のIgA、IgG、IgM抗体価をエライサ(ELISA)法で測定した。その結果、PAcに特異的な血清IgGや唾液IgA抗体価が認められた。一方、pac遺伝子をもたない乳酸菌での経口免疫では、これらの抗体価は誘導されなかった。ついで、組換えプラスミドpSMlで元々PAcを産生しないS.mutans GSーS株を形質転換して、大量のPAcを産生する組換え株を得た。この株が産生する組換えPAcをコレラトキシンBサブユニットと一諸にBALB/cマウスの鼻から免疫したところ、PAcに対するきわめて高い血清IgGや唾液IgA抗体価が誘導できた。そこで、このようにして免疫したBALB/cマウスの口腔内にS.mutans MT8148株の生菌体を5日間毎日接種したのち2週間後にS.mutansを歯面から回収した。その結果、組換えPAcとコレトキシンBサブユニットで免疫したマウスの口腔内からはS.mutansがほとんど回収できなかった。一方、非免疫群やコラトキシBサブユニット単独免疫群のマウスからは多数のS.mutansが回収された。以上の結果から、PAcを発現する乳酸菌による経口免疫や組換えPAcによる経鼻免疫で齲蝕が予防できる可能性が示唆された。
