人工基質小胞の開発

研究概要

軟骨・骨の石灰化には、基質小胞と呼ばれる生体二重膜で囲まれた小胞構造が重要な役割を果していると考えられている。しかし、基質小胞がCaとPiを取り込む機構は不明である。
1.本年度は、軟骨細胞培養系の培地より分離した基質小胞から精製したアルカリホスファタ-ゼと骨より精製した酵素に対する抗体との交差性を検討して、基質小胞アルカリホスファタ-ゼが組織非特異的型(骨型)のアイソザイムであることを確認した。
2.また、II型コラ-ゲンが軟骨細胞培養系から分離した基質小胞に結合していることを見いだした。さらに、軟骨細胞培養系の基質形成期および基質石灰化期より細胞外基質を分離してラット及びモルモットに注射して、それぞれに対する抗体を作成した。
3.さらにこれらの1gGを結合した抗体カラムを作成した。基質形成期の細胞外基質に対する抗体のカラムに、基質石灰化期の細胞外基質成分を通して、石灰化期に特異的に出現するタンパク質を分離した(日本骨代謝学会、1990発表予定)。
現在、石灰化関連蛋白質に対するモノクロナ-ル抗体を作成している。これらの石灰化蛋白質が精製できれば、アルカリホスファタ-ゼとコラ-ゲンとともにリポソ-ムに組み込むことにより、機能的な人工基質小胞を作成することができると期待できる。
