研究概要

非常に少数のミオシン頭部と相互作用している一本のアクチンフィラメントの発生する力をサブピコニュ-メント、サブミリ秒の超高分解能で測定することのできるマイクロニ-ドル操作法を開発して、化学一力学共役の素過程の特性を直接反映する力のゆらぎの解析を行なった。その結果、等尺条件下ではATP分解反応に帰結することのできる大きな力のゆらぎが観察され、コンピュ-タ-シュミレ-ションによる解析の結果、ミオシン頭部は力の発生と非発生の2つの状態を確率的に独立的にとるランダムプロセスで説明できることが明らかになった。これに対して、アクチンフィラメントが滑走しているときは力のゆらぎはほとんど消失し、滑走運動中のミオシン頭部はATPサイクル中ほとんど一定の小さな力を出していることが明らかになった。すなわち、アクトミオシンは負荷に応じて化学一力学共役を変調し、非常に巧妙にエネルギ-変換していることが明らかになった。
