Research Abstract

細胞内伝達分子MAPKの一つであるextracellular signal regulated kinases (ERK)のリン酸化を用いて,末梢からの様々な刺激による後根神経節ニューロンの興奮性の変化を検討してきた.本研究では、関節炎モデルを作成し、本法を応用することで、関節など深部組織の炎症による後根神経節の神経ニューロンの興奮性変化を明らかにする。特に皮膚組織からの入力との相違に注意を払って解析し、深部組織特異的は炎症性疼痛のメカニズムを明らかにすることが目的であった。
前年度までの実験で、ラットの膝関節に炎症惹起物質を投与し関節炎を作成した。ラットの膝関節内へのComplete Freund's adjuvant (CFA)やkaolinとカラゲニンの投与により炎症を起こし、その状態を関節周囲径や関節組織の組織学的評価により評価し、モデルとして使用可能であることが確認できた。フルオロゴールドを投与し、膝関節を支配する後根神経節を同定することが可能となった。
今年度の実験により、
(1)脊髄腔内、もしくは膝関節内に、ERKの上流のkinaseの阻害剤を投与し、一次知覚ニューロンにおける膝関節屈伸運動における、ERKのリン酸化の変化が抑制できることが確認できた.
(2)脊髄腔内、もしくは膝関節内に、ERKの上流のkinaseの阻害剤を投与し、一次知覚ニューロンにおける膝関節屈伸運動における可動域の変化が改善することが明らかとなった.
(3)腫脹した膝関節の屈伸運動によるpERKの発現と、ATP受容体であるP2X3が高い共存率を示すことが明らかとなった.
以上の結果は,ERKを用いた新しい関節痛の評価が確立されたこと,細胞内情報伝達系の亢進が関節痛のメカニズムに重要な意義を持ち,その阻害で疼痛が軽減したことより,全く新しい痛み軽減の戦略のシーズが解明されたと言える.
