Research Abstract

1. 流行地域住民の血清の反応性を調べたところ、ほとんどの抗体がSE36抗原内の8mer反復アミノ酸配列及びそれに続くSer rich配列にのみ反応することから、この領域が防御エピトープであることが強く示唆された。また、リスザルによる免疫-感染実験を詳細に検討したところ、防御エピトープに対する抗体の誘導はSE36ワクチン接種には、ワクチン接種と感染の両方が重要であることが示唆された。そこでTLR9リガンドをアジュバントとして添加し、ワクチン接種のみで効果的に防御エピトープに対する高い抗体価を誘導可能な第二世代SE36マラリアワクチンの開発を進めている。
2. P.vivax ATCC 10株のsera遺伝子の多型を解析した。P.vivaxでは、Pvsera4が、ワクチン抗原Pfsera5のカウンターパートであることが推測されているが、Pvsera4の塩基多様度πは0.04609であり、Pfsera5(π=0.00021)よりも約200倍大きく、多型の頻度が高いことで知られるPvmsp1(π=0.0238)、Pfmsp1(π=0.00299)やPvcsp(π=0.00406)、Pfcsp(π=0.01059)よりもさらに大きい値が示された。今回の解析から、Pfseraに比較して、Pvseraは哺乳類宿主から大きな免疫選択圧を受けてきた可能性が示唆された。
3. 世界各地の原虫集団について熱帯熱マラリア原虫(P.falciparum)のハウスキーピング遺伝子(serca, adsl)、抗原遺伝子(mspl, csp)のシーケンスを得、遺伝的多様性を調べた結果、ハウスキーピング遺伝子ではアフリカが最大で、アフリカからの地理的距離に応じて低下していた。地域原虫集団の起源年代の推定結果と合わせると、P.falciparumはアフリカにおいて現生人類に古くから感染しており、ヒト集団とともに約6万年前にアフリカから世界に広まったことが示唆された。さらに、原虫集団の多様性がマラリア伝播度、媒介蚊の種、過去のマラリア対策によって大きな影響を受けていないことも示唆された。
4. 三日熱マラリア原虫(R.vivax)は系統的にマカク類サルマラリア原虫に近縁であるが、サルには感染しないと言われている。これが本当かどうかを検討するために、各種サルマラリアに高い感受性を示すニホンザルに対してP.vivaxの感染実験を試みた。結果、調べたP.vivax7株はすべて感染しなかった。
