Research Abstract

本研究課題では、これまでに開発を進めてきた新しい光学顕微鏡技術を用い、膜超分子モーター(主にFoF1-ATPase)の作動原理について、1分子のレベルで解明することを目的とする。研究代表者の代表的な特許として、(1)粒子の運動を3次元で検出することのできる光学系、および(2)色素1分子の角度を高い精度で決定できる全反射型顕微鏡がある。これらの技術について、空間分解能と時間分解能の2点について性能を向上させ、光学顕微鏡の新しい手法を切り開く。そしてこの手法を通じて、蛋白質の内部における特定の部位の構造変化の検出に応用する。ダイナミックな構造変化が期待される部位を1個の蛍光色素で標識し、その部位のわずかな動きが定量的に捉える。従来はNMRやX線結晶解析でしか議論されなかった蛋白質の構造変化について、活性を持った1分子を対象に研究を行うことができるようになるはずである。
