Outline of Annual Research Achievements

本研究ではケモテクノロジーを用いて5年間の研究期間中にユビキチン修飾の多様な機能を解析することを目指す。最終年度である本年度は以下の項目から研究を推進した。
1. M1鎖を形成するLUBACユビキチンリガーゼはHOIP、HOIL-1L、SHARPINの3サブユニットから構成され、HOIPがM1鎖を形成する活性中心を持つ。HOIL-1Lもユビキチンリガーゼ活性を有しており、研究代表者らはHOIL-1Lのユビキチンリガーゼ活性を欠失させることでLUBACのM1鎖形成能を亢進させて細胞内感染菌の感染を抑制することを見出した。そこで、LUBAC機能亢進剤の感染症治療としてのポテンシャルを検討すべく、HOIL-1Lリガーゼ(E3)欠失マウスを用いてサルモネラ感染実験を行った。しかし、HOIL-1L E3欠失マウスは正常マウスよりもサルモネラ感染が増悪したので、開発を中止した。

3.ユビキチン修飾の新機能のケモテクノロジー解析:昨年度に同定した細胞内でもM1鎖とHOIL-1L NZFの結合を阻害2種の化合物のうち、1つはNF-kappaB活性化を抑制するが、細胞死を抑制しないこと、もう1つは細胞死を亢進させることを示した。2つの化合物が異なる特徴を示すのは細胞死を亢進させる化合物はHOIL-1LのNZFだけではなく、SHARPINのNZFとM1鎖の結合を阻害するためであることを示した。これらの化合物を用いた解析からHOIL-1LのNZFは主にNF-kappaB活性化に、SHARPINのNAFは主に細胞死抑制に機能することが明確となった。現在、論文投稿準備中である。
