オピオイド受容体の網羅的シグナル解析による薬理経路の同定

Outline of Annual Research Achievements

本研究課題の対象GPCRであるカッパーオピオイド受容体(KOR)に対するリガンドとして斉藤班が設計・有機合成した12種類のYNT-1612類縁化合物について、KORを介したGタンパク質活性(GiおよびGo)とアレスチン活性をNanoBiT-GPCRシグナルアッセイを用いて評価した。その結果、元化合物のGタンパク質バイアスリガンドであるYNT-1612と比較して、Gタンパク質活性のEfficacyが同等かつアレスチン活性のEfficacyが減少し、Potencyが増加する類縁化合物を複数見出した。また、デルタオピオイド受容体(DOR)に対するリガンドとして斉藤班が設計・有機合成した7種類のKNT-127類縁化合物について、同様の解析を実施したところ、KNR-127とGタンパク質活性のEfficacyが同等でアレスチン活性のEfficacyが減少した類縁化合物を複数見出した。一方、これらの化合物はPotencyがKNT-127よりも低かったことから、有用性は低いものと判断した。
寿野班が構造解析を実施したKORについて、リガンド間で構造に差が見られた部位について変異体実験を行った。その結果、KORについて、アレスチン結合のみに重要なアミノ酸残基を2箇所同定した。DORについても同様に変異体実験を進め、クライオ電顕解析で得られたリガンド結合様式を検証した。
斉藤班が個体の薬理実験に使用した7種類のオピオイドリガンドについて、ヒトとマウスのオピオイド受容体に対するGタンパク質活性とアレスチン活性を網羅的に計測した。その結果、多くのリガンドの挙動は既報に一致したオピオイド受容体サブタイプの選択性を示すことを確認するとともに、一部のリガンドが広範のオピオイド受容体サブタイプを活性化させるパーシャルGタンパク質バイアス型であることを見出した。
