Outline of Annual Research Achievements

本研究では、金属酵素のストラテジー (戦略)を触媒開発に応用し、効率的かつ位置および立体選択的な脂肪族および芳香族化合物の水酸化反応の開発をめざして研究を行った。(1)新規な単核銅オキソ錯体の開発:銅を含む一原子酸素添加酵素の中には、単核銅活性中心において分子状酸素を還元的に活性化し、各種有機基質の酸素化反応を効率よく達成しているものがある。本年度は、このような金属酵素活性中心に含まれる単核銅活性酸素種のモデル錯体の創成をめざして検討を行い、これまでに殆ど報告例のない単核の銅(II)スーパーオキソ錯体や、これまでに全く報告例のない単核の銅(II)オキシラジカルの創成に成功した。用いる配位子としては、後藤 敬教授(東工大院理工)らの開発した嵩高い置換基を導入した新規なボール型配位子を合成した。また、同様の配位子を用いてニッケル活性酸素錯体の創成にも成功した。(2)新規なオスミウム錯体を用いたオレフィンの触媒的酸化反応の開発:オスミウムを錯体化することにより反応性や反応の選択性を制御し、環境に優しく、より効率的な酸化触媒の開発をめざして検討を行った。その結果、オレフィンのジオール化やアミノヒドロキシル化反応における効率的な触媒として機能することを見いだした。また、森聖治教授(茨城大理)との共同研究により、DFT計算を用いた反応機構の解明にも成功した。(3)人工金属酵素の創成:単核や二核の銅および鉄活性中心を有する人工金属酵素の創成をめざして、高熱菌由来のタンパク質を鋳型とする金属錯体の調製を行った。得られた金属タンパク質の結晶構造や分光学的特性を明らかにすると共に、基本的な酸化反応性を明らかにした。
