Research Abstract

脱ユビキチン化酵素A20 がC 末端のzincfinger 7(ZF7)を介して直鎖状ポリユビキチンに結合することでNF-κB 活性化を抑制することを,生化学的な解析,および,細胞生物学的な解析により明らかにした.A20 ZF7-直鎖状ユビキチン複合体のX 線結晶構造を決定し,その直鎖状ジユビキチン認識機構を明らかにした.構造情報に基づいて機能解析を行い,A20 がZF7 を介して直鎖状ポリユビキチンと結合しTNF 受容体へと集積することでNF-κB 経路を制御するという新たな分子機構を明らかにした.さらに,本研究の結果から,A20 ZF7と直鎖状ポリユビキチンとのあいだの結合の不全はB 細胞リンパ腫惹起と関連することが示された.

Reason

A20に関しては,結晶構造解析だけではなく,領域内の異分野連携により細胞生物学的解析,分子生物学的解析を行い,NF-κB 経路を制御するという新たな分子機構を明らかにできた点では,順調に当初の目的が達成できていると言える.

Strategy for Future Research Activity

HTLV-Tax蛋白質,A20などのNF-kB経路,MAPK経路等に関わる因子の構造解析を,計画班間との共同研究により遂行する.HTLV-Tax蛋白質に関しては, NF-kB経路の因子であるNEMOとの複合体含め,複合体の構造解析をすすめている.その他,現在進行中の領域内の計画班・公募班との共同研究を進めるだけでなく,新たな共同研究も展開する.
