Outline of Final Research Achievements

Genome adaptation during meiosis promotes new combination of genetic information in next generation. A DNA exchange reaction, homologous recombination plays a critical role in genome adaption. However, it still remains how recombination during meiosis is controlled at a molecular levels and how chromosome dynamics regulate the reaction. In this study, we found : 1. Transcription coupling epigenetic marks regulates the initiation of the meiotic recombination. 2. Post-translational modification of the proteins in nuclear envelope regulates the recombination through control of chromosome dynamics. 3 Crystal structural analysis of the protein complex involved in meiotic recombination provide a novel insight on the mechanism of the homology search in the recombination. Given the dysfunction of genome adaptation confers genome instability in gametes, which are often associated with miscarriage and Down syndrome, our knowledge might contribute prevention of age-dependnet miscarriage in human.
