Research Abstract

我々は、今まで造血幹細胞を中心に細胞周期の静止期性、代謝の特徴、幹細胞ニッチの低酸素性について研究を進めてきた。後半では癌幹細胞研究により集中し、慢性骨髄性白血病(CML)と骨髄腫の解析を進め、ニッチ療法の開発をめざしてきた。CMLは、造血幹細胞異常による骨髄増殖性疾患である。我々はマウス造血幹細胞にbcr/ablを導入し、それらをマウスに移植してCMLモデルを作成し、以下のように、腫瘍由来細胞がニッチとして作用していることを示した ( Blood, in press)。
1)CML幹細胞はKSL分画に存在し、その中でもCD25(IL-2Rα)陽性細胞が、Leukemia-initiating cellである。
2)CD25(IL-2Rα)KSL陽性細胞はヒトCML患者にも認められる。
3)CD25(IL-2Rα)KSL陽性細胞は、IL-6、IL-4などTH2サイトカインを多く発現していること、それらのサイトカインが血中で上昇している。
4)CD25(IL-2Rα)KSL陽性細胞を抑制する抗体をマウスに投与したところ、CMLの進展を抑制しうる。同様にCD25欠失マウスにおいても、CML進展の抑制をみた。

Reason

マウスCMLモデルにおいて、白血病幹細胞およびニッチ細胞が産生するサイトカインが、白血病の病態に大きくかかわることを明らかにすることができた。

Strategy for Future Research Activity

i)マウスCMLモデルにおいて同定したCD25分子が、ヒトCMLでも病態に関与するか否か検討を加えたい。
ii) 多発性骨髄腫(MM)は、B細胞由来腫瘍細胞が主として骨髄内で増殖する悪性腫瘍である。GFP標識したヒト骨髄腫細胞を免疫不全マウスに静脈から投与し、MMを作成するというモデルをすでに作成している。このモデルを用いて、MMでは、どのような機構によって正常造血が抑制されるかを明らかにする。
