Outline of Annual Research Achievements

我々はマウスCML modelにおいて、CD25(IL-2Ra) 陽性細胞が、IL-6 やIL-13のような炎症性サイトカインを産生し、CML細胞の増殖を促していることを見出した。また、IL-2Rシグナルを遮断することにより、この炎症性ニッチを解除できることから、CML治療に有効と考えた(Blood, 2014)。ヒトCMLにおいても、CD25陽性細胞は検出でき、現在、慶應病院内科において臨床試験の可能性を探索している。
また、我々は、p53関連遺伝子であるASPP-1の欠損マウスの解析を行い、造血幹細胞のapoptosis が抑制され、造血再建能など幹細胞機能が高いことを認めた。しかし、本マウスは造血幹細胞においてDNA 損傷fociが優位に高く、p53とASPP-1 の二重欠損マウスにおいて、T-cell leukemia/lymphomaの発症頻度が優位に高いことを認めた。ヒトALLのうち、ASPP-1のメチル化された症例は予後不良であることが報告されており、DNA損傷と白血病発症の関係に新しい視点をもたらしたと考える (in revise)。
