Outline of Annual Research Achievements

タンパク質ライブラリーから目的タンパク質を単一工程で単離･純化する半田ビ－ズ技術により、生物活性を持つ天然物リガンドとの特異的結合タンパク質を単離･同定し、その中からリガンドの活性に関わるターゲットを決める一般的なスキーム(1)~(5)を確立し、そのシステムの有効性を検証する。(1)半田ビーズへ天然物リガンドの固定化技術、(2)タンパク質ライブラリー調整法、(3)アフィニティ精製による結合タンパク質の単離･同定技術、(4)結合タンパク質からターゲット同定技術、(5)ターゲット関連ネットワークの解析法、を確立し、本研究班の発展に貢献すると共に、本システムによる具体的成果を挙げる。成果として、①グルタミン合成酵素が細胞内のグルタミン濃度を維持するメカニズムを解明した。グルタミンが高濃度になると11番と14番のリジンがアセチル化され、分解シグナルの「デグロン」構造を呈し、CRBN E3ユビキチンリガ－ゼと結合し、Ub化･分解されることを明らかにした（Mol. Cell, 2016）。②Progestrone receptor membrane component 1はヘムと結合し、ヘムを介して2量体化するとEGFRやCYP450と相互作用し、がん細胞の増殖シグナルや異物代謝を亢進することを示した。COはヘムの疎水界面に入り込み、PGRMC1の2量体化を阻害し、増殖シグナルなどを抑制することを示した（Nat. Commun., 2016）。③サリドマイド/ターゲットのヒトセレブロン（CRBN）/会合因子のDamage-specific DNA-binding protein 1複合体のX線結晶構造を解明し、サリドマイドがCRBNのC末端と結合し、光学異体S体がR体より安定により強く結合することを示した（Nat. Struct. Mol. Biol., 2014）。
