Research Abstract

1. 紅色細菌由来のLH2とLH1-RCの機能解明:紅色光合成細菌Blc. viridisからLH1-RCコア複合体を、Rps. acidophilaからLH2複合体を単離し、これら色素タンパク複合体を脂質二重層膜に任意の割合で組み込んだ光合成再構成膜(人工光合成膜)を構築し、その超分子構造はAFMで、その機能はフェムト秒時間分解共鳴誘導ラマン分光法で解明した。
2. 緑色細菌由来のクロロゾームの超分子構造・機能解明:我々が開発したバクテリオクロロフィルeを生産する緑色光合成細菌の遺伝子改変法を利用して、そのC7位のホルミル化酵素遺伝子bciDやC20位のメチル基転移酵素BchUのいずれか、もしくは両方を欠損させた変異体を調製し、バクテリオクロロフィルcやf、もしくはdのみを生産する菌体を作成し、その主たるアンテナ器官であるクロロゾームを単離精製した。天然体を含む4種のクロロゾームにおける光励起エネルギー移動過程を明らかにした。
3. 光合成アンテナとメソポーラス物質の連結体の創製:水の酸化触媒であるイリジウム錯体[Ir(bpy)Cp*Cl] をビピリジン-メソポーラス有機シリカ(BPy-PMO)に固定したところ、均一系と同等の活性発現と耐久性向上を確認できた。金属錯体は一般に耐久性が低く実用化の最大のネックの一つになっていたので、実用化に向けたネックの一つを解決する足掛かりが得られた。

2: Research has progressed on the whole more than it was originally planned.

Reason

1)紅色細菌由来のLH2とLH1-RCの機能解明は、予定通りの進展がみられた。2)緑色細菌由来のクロロゾームの超分子構造・機能解明は、様々な緑色光合成細菌の遺伝子改変体の調製に成功し、機能解明の面で予想以上の進展が見られた。3)光合成アンテナとメソポーラス物質の連結体の創製は、人工光合成系の実用化に向けて大きく発展しており、達成度は及第点と考えている。

Strategy for Future Research Activity

これまでの研究成果を基にして、さらに1)褐藻や珪藻由来のFCPと紅色細菌由来のLH1-RCの機能解明と2)緑色細菌由来のクロロゾームの超分子構造・機能解明と人工光合成アンテナの創製を行いつつ、3)光合成アンテナとメソポーラス物質の連結体の創製の創製を進めていく。
