Outline of Annual Research Achievements

この計画班ではタウタンパク質の生理機能が加齢に伴い神経変性を引き起こす凝集体形成を行うまでの分子機構を明らかにすることを目的にしている。
タウリン酸化に関して霊長類モデルとして使用されているマーモセットの内在性タウについて調べた。リン酸化に関して生後直後に増大し成体になるとリン酸化タウの減少が観察された。タウの配列、アイソフォームの観察からマーモセットには霊長類特有の配列が欠損し、アイソフォーム変化も齧歯類と同様の変化を示した。
ヒト脳では生体においても4R,3Rタウのアイソフォームが存在している。これらタウアイソフォームの役割を調べたところ4Rタウノックインのホモマウスにおいて、有意に成体神経新生細胞数が減少することを見出した。またタウノックアウトマウスでは新生細胞の移動距離が増加しており、このことはタウが微小管の安定化を促進することより、タウの喪失によって新生神経細胞の移動性が増加することを反映していると推測された。
タウの分布についての検討ではタウは主に無髄線維およびオリゴデンドロサイトに局在し、タウ病変が好発する錐体細胞では相対的なタウの発現量は低かった。タウの発現制御機構について転写因子としてSOX11/12 を同定した。転写によって生成されたタウmRNAはRNA結合タンパク質と複合体を作り細胞体、樹状突起に存在している。神経過活動によりmRNAからの翻訳活性化が起こり、後シナプス領域でタウ発現が起こる。このタウの役割を調べるためグルタミン酸刺激後のAMPA受容体の1分子観察を行った。その結果、グルタミン酸刺激によってAMPA受容体の移動速度は増大するがタウがないと細胞内への取り込みが阻害されシナプス外でのAMPA受容体の移動が阻害されることが明らかになった。このことからシナプスにおけるタウの増大はAMAP受容体を取り込み神経過活動を抑制する補償機構として作用し得ることが示唆された。
