真核細胞における鞭毛・繊毛運動の分子機構の解明

Research Abstract

滑り運動の分子機構に関して高橋は独自に開発した装置で、高Ca^<2+>がウニ精子鞭毛の特定の滑りパタ-ンを高頻度で起すことを示し、上村は10^<ー9>mの精度でウニ精子鞭毛の微小管滑り運動を解析することに成功し、滑りがチュ-ブリンのサイズを反映した振幅で起ることを発見した。神谷はダイニン標品上を微小管を滑らせる実験系を開発し、ダイニンは異なる運動性を起す複数部分からなることを示し、豊島は同様の系で、ダイニンATPaseはキネシン、ミオシンのATPaseに比べ基質特異性が高いこと、加水分解と運動の速度の相関は低いことを示した。毛利はトリプシン分解法でウニの外腕ダイニンβ鎖のATP分解に伴うconformation変化とα鎖の相互作用が滑り運動に重要であることを示し、中村は微小管と微小管結合タンパクの給合を解析し、チュ-ブリンサブユニット間の数個の架橋様式の存在を示した。繊毛運動に関して馬場は、ウニ幼生繊毛に対する神経伝達物質の作用を調べ周期運動のONーOFFと対称性を調節している因子について示し、中岡はゾウリムシで細胞膜の電流変化が繊毛打頻度を調節している事を明らかにした。野口はゾウリムシ繊毛打の逆転がCa^<2+>の膜での流入流出により調節されていることを示した。精子運動開始の分子機構に関し、林はその中核部分がAキナ-ゼ、チロシンキナ-ゼ、15Kタンパクの複合体からなることを示し、又15Kタンパクの抗体を作製した。森沢は海産魚精子の運動開始には細胞内Ca^<2+>、細胞内pHが重要な役割を果していることを明らかにし、奥野は海産無脊椎動物フジツボでCa^<2+>の関与について研究を行った。渡辺はテトラヒメナのカルモジュリンファミリ-に属する3種のCa^<2+>給合タンパクを発見し、更にこれらの遺伝子のクロ-ニングとシ-クエンシングに成功した。又、これらの遺伝子のアンチセンスmRNAを調製し、微量注射法で、これらの結合タンパクの繊毛運動における機能について検討中である。
