Research Abstract |
DNA相同組換えは、減数分裂期にゲノムに多様性をもたらすのみならず、様々な要因によって生じるDNA損傷を修復する重要な生物機能の1つであるRad51は組換え反応を触媒し、大腸菌から人まで高度に保存されている重要なタンパク質である。これまでに私は、分裂酵母において接合型変換、DNA修復、減数分裂期組換えに関わるswi5遺伝子の解析を行い、Rad51 (Rhp51)依存的組換え修復経路において、Swi5依存的経路を新しく同定した。また、Swi5が接合型変換時にはSwi2と、DNA修復時にはSfr1と複合体を形成し、Rad51 (Rhp51)による組換え反応を促進していることを明らかにした。この結果を、2003年12月23日号のProceedings of the National Academy of Sciences of the United States of Americaに発表した。 さらに、Swi5の機能を詳細に調べるために、細胞内でSwi5-EGFP融合タンパク質を発現し、細胞内局在を観察した。Swi5-EGFPは核に局在し、さらに核内で数個のfociを形成していた。Heterochromatin protein 1 (HP1)の分裂酵母ホモログであるSwi6をECFPと融合し、Swi5-EGFPとの共局在を調べたところ、Swi5-EGFPとSwi6-ECFPが共局在し、Swi5がヘテロクロマチン領域に局在することが分かった。これまでの実験結果から、Swi5がSwi2を介してSwi6と相互作用すること、Swi2-EGFPが核内で数個のfociを形成することが明らかになっている。これらのことから、ヘテロクロマチン領域ではSwi5-Swi2-Swi6の複合体が形成され、何らかの機能を担っていることが予想される。一方、Swi5-EGFPは、紫外線照射後に細胞内で新たにfociを形成した。またSfr1-EGFP融合タンパク質も紫外線照射後に細胞内でfociを形成した。多くの相同組換えに関わるタンパク質についてDNA損傷部位に結合することが報告されているため、Swi5がSfr1と共にDNA損傷部位に局在している可能性が考えられる。
