Research Abstract	本研究班では癌化学療法を発展させるために新しい作用および構造を有する制癌剤の発見を目的に、進展の著しい研究領域の中から情報伝達因子、細胞増殖制御因子および耐性因子などを標的に有効物質を探索、その作用機作を検討し制癌剤としての可能性を追究している。本年度は以下の研究成果を挙げた。抗生物質azatyrosineは活性化CーHaーras遺伝子で癌化したNIH3T3細胞の増殖を阻害するとともに癌化細胞を正常化する作用を示すが、azatyrosineによって正常化したリバ-タント細胞の遺伝子を解析したところ、その1つは抑制遺伝子rrgであることを明らかにした。海綿由来の抗腫瘍および抗ウイルス物質mycalamideAおよびBに癌細胞を正常化させる作用を見出した。制癌作用を示すサイトカイン産生誘導物質を微生物代謝産物中に探索し、放線菌の生産する新物質conageninおよびcytogeninを発見した。conageninはT細胞に作用しILー2およびILー3の産生を増強し、cytogeninはマクロファ-ジによるILー1産生誘導作用を示し、いずれの物質もマウス固型腫瘍の増殖を抑制した。TNF産生に作用する物質を得ることを目的にソルビト-ルにオレイン酸を結合させた物質を合成したところ、マウス固型腫瘍に制癌活性を示した。本物質のTNF産生に対する作用は検討中である。制癌抗生物質15ーdeoxyspergualinが血管新生抑制作用を示すこをを明らかにした。多剤耐性因子gp170を標的として得られたジハイドロピリジン系耐性克服物質はvincristine(VCR)耐性P388白血病マウスに対しVCRとの併用により著明な延命効果を示した。新規抗生物質、resorthiomycinは多剤耐性株に親株より低濃度で増殖阻害効果を示した。合成イソプレノイド(SDBーethylenediamine)は制癌物質の細胞内への取り込みを増強し、細胞内濃度を上昇させる新しい耐性克服物質であることを明らかにした。
