Research Abstract

本総合研究では、昆虫後期発生過程に特徴的な諸生命現象を理解するため、1)遺伝子レベルで解析の進んでいる卵黄タンパクおよび貯蔵タンパク生合成系、2)生体防御に中心的役割を有するフェノ-ルオキシダ-ゼ系、3)組織崩壞と新生に関与する血球細胞系、4)昆虫飛翔行動のエネルギ-供給系としてのリポホリンーアポリポホリン系、を対象として遺伝子・タンパク質レベルでの解析を進め以下の成果を得た。
1)カイコおよびイエバエ脂肪体cDNAライブラリ-よりビテロゲニンcDNAクロ-ンを単離、塩基配列を決定した。RNAブロット解析によりビテロゲニン生合成の性ならびに発生段階特異性はmRNAレベルで制御されていることを証明した。カイコ卵特異的タンパク質遺伝子を単離し、全塩基配列を解明した。ビテロゲニン分解システインプロテア-ゼを単離・精製しN末端領域のアミノ酸配列を決定した。カイコ脂肪体細胞膜より貯蔵タンパク質とり込み系を確立した。
2)カイコ体液よりフェノ-ルオキシダ-ゼ活性化カスケ-ド系を分離、再構成した。カイコ外皮ラッカ-ゼおよびショウジョウバエ・フェノ-ルオキシダ-ゼ前駆体の活性化にタンパク分解酵素の関与を証明した。
3)センチニクバエ蛹化にともなう脂肪体の崩壞には血球細胞表面に出現する200KDaが関与し、このタンパク質と脂肪体細胞の結合により血球細胞よりキモトリプシン様プロテア-ゼが放出され脂肪体を分解することを確認した。
4)トノサマバッタ低密度リポホリン(LDDP)に結合しているアポリポホリンIIIの量は含有ジアシルグリセロ-ル量に依存して変化し、LDDP1分子に約9分子のアポリポホリンIIIが結合していることが判明した。
