先天性免疫不全症成立におけるIg遺伝子プロモ-タ-結合たん白の意義に関する研究

Research Abstract

Ig遺伝子プロモ-タ-結合たん白であるOCT2たんぱくのmRNAの発現をみるためにreverse transcriptase Polymerase Chain Reaction(RTーPCR)法を開発した。種々の造血細胞でこの遺伝子の発現を解析したところ、リンパ系の細胞における強い発現をみた。この発現がfms遺伝子導入ーCSFー1刺激B細胞でも同様に認められるかいなかを検討するためにcーfms遺伝子のレトロウィルスを作製した。cーfms遺伝子(Dr.Sherrより供与)cDNAをPzip SV(X)neoベクタ-のBamHIサイトに導入し、パッケ-ジング細胞株である4cripにトランスフェクトした。cーfmsレトロウィルスのタイタ-を測定する為にヒト上皮細胞株であるKM13に導入し、neomycineで選拓したところ1×10^3/mlのウィルスタイタ-を得た。ヒト単球細胞株であるU937細胞にこのウィルスを感染させ、neomycineにより選拓することによってcーfms導入U937株を10株作製した。この細胞を60U/mlのCSF1(Immunexより供与)存在下で培養することによってcーfmsを介するシグナルがU937細胞をマクロファ-ジに分化させることを確認した。TPA処理によってマクロファ-ジへと誘導されたU937ではcーfmsの発現がPCRによっても確認できないことから、TPAを介する径路とは異った径路の役割りがcーfmsの径路には存在すると考えられた。現在このU937におけるOCT2 mRNAの発現の変化を解析すると同時に、EBウィルス形質転換B細胞にcーfmsを導入する計画を実施中である。これらの系におけるOCT2 mRNAの変化を解析してみたい。又先天性免疫不全症患者リンパ球については患者検体を収集中である。
