Research Abstract |
1.発色団周辺残基すべてをBRと同一に変異させた多重変異体BR/ppRによる解析 BR/ppRの極大吸収波長(λmax)は、524nmでありBRのそれと同じとはならず、また赤外分光の結果、発色団プロトン化シッフ塩基(PSB)の水素結合力は野生型と同じく強いままであることがわかった[Shimono et al.(2001)BBA 1515,92-100;Shimono et al.(2002)Biochemistry 41,41,6504-6509]。これらは、ppRのPSB周辺が、BRとは微妙に異なることを示し、この違いは発色団周辺のアミノ酸側鎖の違いよりも遠距離相互作用によって決定されていると考えられる。このような違いが吸収波長制御の主要因であると考えられる。 2.キメラ蛋白質を用いた遠距離相互作用の解析 PSB(ヘリックスG)とその対イオン(ヘリックスC)の相対配置を変化させる目的で、2種のキメラ蛋白質を作製した(B-ABC/P-DEFGとP-ABC/B-DEFG)。ここでB-ABC/P-DEFGは、ヘリックスABCがBRの配列で、ヘリックスDEFGがppRの配列を持つキメラ蛋白質である。B-ABC/P-DEFGとP-ABC/B-DEFGのλmaxは、それぞれ506,545nmであった(BRのλmaxは、550nm)。これらは、ヘリックスD以降に吸収波長制御因子が存在することを示す。次にこの因子の存在範囲を限定するために、すべての組み合わせのキメラ蛋白質を作製し、そのλmaxを比較した。その結果、ヘリックスDとEの相互作用による効果とThr204(ヘリックスG)の効果が、BRとppRの吸収波長の違いを引き起こす主要因であることがわかった[Shimono et al.J.Biol.Chem.審査中]。
