Research Abstract |
海馬や大脳新皮質の錐体細胞の樹状突起は興奮性・抑制性シナプス入力が統合される重要な部位である。我々は本研究において、錐体細胞の樹状突起におけるシナプス統合や可塑性の仕組みを、樹状突起遠位-近位軸に沿って詳細に解明することを試みてきた。特に本年度は、強いシナプス刺激により誘発される海馬錐体細胞の同期的振動現象(後発射)は興奮性GABA作動性入力によって駆動されることを見出したため(Tsukamoto et al.2003)、錐体細胞のどの局所部位(尖頭樹状突起、基底樹状突起、あるいは細胞体)がこのような興奮性GABA伝達を担っているのかに焦点を当てた研究をおこなった。まず、クロライド・イメージング法をもちいて、GABA受容体の逆転電位を決定する塩化物イオンが後発射誘発中に錐体細胞の細胞体内に蓄積することを観察し(Isomura et al.2003c)、海馬の同一領域内における後発射の発生にはGABA伝達が関与する一方、隣接領域への後発射の伝播にはグルタミン酸伝達が関与していることも示した(Isomura et al.2003b)。さらに、錐体細胞と介在細胞の後発射の同時記録を樹状突起遠位-近位軸に沿って解析したところ、両細胞群が基底樹状突起領域内に特異的に形成する正の帰還回路が後発射の同期現象の本質的なメカニズムであることを示唆する結果を得た(Tsukamoto et al.印刷中)。このように、海馬錐体細胞における同期的振動現象の発生には、細胞体や基底樹状突起への興奮性GABA入力が重要な役割を果たしていることを示す知見を得ることができた。
