Research Abstract |
当研究室において、ファイトプラズマの昆虫伝搬能喪失変異株(OY-NIM)の作出に成功し、接木により維持している。このことはファイトプラズマが存続し続けるためには宿主植物-媒介昆虫間のホストスイッチングが必須ではないことを示唆している。本研究では、これまでの結果をもとにファイトプラズマのホストスイッチングに関わる遺伝子を同定し、発現の比較解析を行うことを目的としている。まず、野生株(OY-W)と比較してOY-NIMの染色体では130キロベースにおよぶ欠失と、染色体外DNAにおいていくつかのORFが欠失していることを明らかにした。染色体における欠失領域の解明は現在進行中であるが、染色体外DNAにおいて欠失しているORFの解析を行ったところ、欠失しているORFは2つあり、1つは1本鎖DNA結合タンパク質をコードする遺伝子で、もう1つは機能未知のタンパク質をコードしていた。後者を機能予測プログラムを用いて解析したところ、2ヶ所の膜貫通領域と、膜輸送系の1つであるSecシステムにより輸送されるためのシグナル配列を持つことが明らかとなり、このタンパク質は膜タンパク質である可能性が高いことが明らかとなった。ファイトプラズマは細胞壁を持たないために膜タンパク質が宿主細胞質に露出しており、宿主タンパク質と直接相互作用すると考えられる。また、スピロプラズマにおいては接着性の膜タンパク質が昆虫伝搬に関わることが知られており、この膜タンパク質がホストスイッチングに関わる可能性が高いと考えられた。
