ALS原因遺伝子が誘導する神経細胞死を抑制する新規因子の解析

Research Abstract |
我々は優性遺伝性筋萎縮性側索硬化症(ALS)の原因遺伝子であるSOD1変異体の発現により誘導される神経細胞死を新規ALS原因遺伝子ALS2がコードする蛋白Alsinが完全に抑制することを見い出した。AlsinはSOD1変異体特異的に神経保護作用を示し、他の神経変性疾患原因遺伝子であるAPP等が誘導する神経細胞死には無効であった。また我々はAlsinの機能ドメインの解析により、この神経保護作用にはAlsin long form(LF)のRhoグアニン交換促進因子(RhoGEF)ドメインが不可欠であることを明らかにした。RhoGEFドメインはDHドメインとPHドメインから構成されている。DHドメインはグアニン交換反応活性を担い、PHドメインはRhoGEFの細胞内での局在に寄与することが知られており、細胞内でGEF活性を示すためには両方のドメインが必要である。AlsinのDHドメイン、PHドメインそれぞれ単独のみでは細胞死を抑制できないことから、SOD1変異体が誘導する神経細胞死に拮抗するにはRhoGEFの活性が重要であることが推測される。さらに、我々はSOD1とAlsinの物理的相互作用を見い出した。野生型SOD1はAlsin short form(SF)と結合するが,Alsin LFとは結合しないのに対し、SOD1変異体はAlsin SF,LF双方共に結合した。我々は結合ドメインを探索し、SOD1変異体がAlsinのRhoGEFドメインと相互作用をもつことを明らかにした。この2つのALS原因遺伝子産物SOD1とAlsinの物理的機能的相互作用の解析はALSの選択的運動神経細胞死のメカニズムの解明において非常に重要であると考えられる。
