Research Abstract

本研究は植物細胞の伸長制御、伸長方向制御、細胞質分裂制御などの機構の解明を目的として行われた。
細胞伸長方向の制御には細胞質表層微小管が関与している。柴岡は、アズキ上胚軸において、ブラシノライドが横方向の微小管を持つ細胞の割合を増加させること、蛋白質リン酸化阻害剤が表層微小管の横方向から縦方向への変換を促進するることを明らかにした。また、植物の細胞質分裂装置である隔膜形成体から隔膜形成体形成に関与していると思われるモーター蛋白質の単離に成功した。園部は、表層微小管の配列制御機構を理解する上で表層微小管付髄蛋白質の性質を知ることが不可欠と考え、タバコ培養細胞から表層微小管付髄蛋白質4種類を単離し、その中の80および65kDa蛋白質の一部アミノ酸配列を明らかにした。馳沢は、表層微小管の配列制御に関与していると考えられる蛋白質のcDNAの全塩基配列を決定し、EF1α様蛋白質であること、精製した蛋白質が微小管重合能を有することを示した。
オーキシンによる細胞伸長は細胞壁中のキシログルカンの分解によるものと考えられる。林は、ポプラ培養細胞を用い、エンド-1,4-β-グルカナーゼのmRNAレベルがオーキシンにより上昇することを見いだした。保尊は、アズキ上胚軸からキシログルカンを分子量数万の大きさに切断する新規エンドグルカナーゼを単離した。西谷は、シロイヌナズナより得たエンド・キシログルカン転移酵素をコードするゲノムDNAのプロモーター領域にジベレリンに反応する配列が存在することを明らかにした。
福田はオーキシンとサイトカイニン存在下で培養する管状要素に分化する単離ヒャクニチソウ葉肉細胞を用い、分化初期に発現するcDNAクローン17個を単離した。
