動脈平滑筋Ca^<2+>活性化K^+チャネルの本態性高血圧症における役割

Research Abstract |
平成5年度はSHRの動脈平滑筋の静止時におけるCa^<2+>活性化K^+チャネル機能と脳動脈と・冠動脈の静止時におけるCa^<2+>活性化K^+チャネル機能を検討し、以下の新しい知見を得た。 1.血圧が十分に高い13週令SHRの頸動脈・大腿動脈・腸間膜動脈の静止時においてはCa^<2+>活性化K^+チャネルが強く機能している。WKYで静止時にこのチャネルは殆ど機能していない。SHRのこれら動脈の静止時には電位依存性Ca^<2+>チャネルが開いており、細胞内へのCa^<2+>流入が増加し、この結果Ca^<2+>活性化K^+チャネルが強く機能し、Ca^<2+>流入による動脈収縮に対し強いフィードバックをかけていると考えられる。このフィードバック機構は非常に重要であり、もしこの機構が破綻するとSHRでは静止時にもかかわらず非常に強い収縮がおきてしまう。 2.SHRの動脈では静止時にも軽度の収縮がみられるので、静止時に流入したCa^<2+>はこのフィードバック機構を強く作動させる同時に、収縮蛋白系にも供給されていると考えられ、このバランスが静止時の張力に大きく寄与している。 3.血圧上昇初期の6週令SHRの頸動脈・大腿動脈の静止時においても同様の現象が見られた。6週令SHRの動脈の静止時におけるCa^<2+>活性化K^+チャネルの機能は13週令SHRに比べやや弱かった。 4.イヌ脳動脈(脳底動脈・後交通動脈・中大脳動脈)の静止時においてCa^<2+>活性化K^+チャネルは強く機能しており、13週令SHR動脈と同様の現象が見られた。このチャネルは冠動脈の静止時においても弱いながら機能していたが、腸間膜動脈では殆ど機能していない。
