動脈平滑筋Ca^<2+>活性化K^+チャネルの本態性高血圧症における役割

Research Abstract |
平成6年度はSHRの動脈平滑筋のCa^<2+>活性化K^+チャネル機能を検討し、以下の新しい知見を得た。 1.血圧上昇前の4週令SHRの大腿動脈の静止時においても、血圧が十分に高い13週令SHRの大腿動脈の静止時(平成5年度に検討)と同様に、WKYに比べCa^<2+>活性化K^+チャネルが強く機能している。4週令SHRの動脈の静止時には電位依存性Ca^<2+>チャネルが開いており、細胞内へのCa^<2+>流入が増加し、この結果Ca^<2+>活性化K^+チャネルが強く機能し、Ca^<2+>流入による動脈収縮に対し強い負のフィードバックをかけている。このフィードバック機構がもし破綻するとSHRでは静止時にもかかわらず非常に強い収縮がおきてしまう。SHRの動脈では静止時にも軽度の収縮がみられるので、静止時に流入したCa^<2+>はこのフィードバック機構を作動させると同時に、収縮蛋白系にも供給されており、このバランスが静止時の張力に大きく寄与している。4週令SHRでみられたこれらの機能は遺伝的素因に関連していると考えられる。 2.内皮由来過分極因子(EDHF)は血管内皮で合成され、種々の刺激に対応して放出され、動脈平滑筋に作用しCa^<2+>活性化K^+チャネルが開口して弛緩がおこる。このように血管には内皮を介したCa^<2+>活性化K^+チャネルによる負のフィールドバック機構が備わっている。SHRの脳底動脈ではEDHFの合成あるいは放出が減少しており、このフィードバック機構が十分機能していない。例えばセロトニンで刺激するとWKYに比べSHRでは強い収縮が認められる。 3.ATP感受性K^+チャネル開口薬の血管弛緩作用をSHR頚動脈で検討したところ、この薬物はATP感受性K^+チャネルを開口して過分極をおこすため、二次的に電位依存性Ca^<2+>チャネルが閉じ、更にCa^<2+>活性化K^+チャネルが閉じる、という複雑な作用を示した。
