Research Abstract

研究目的:本研究の目的は、(1)血中免疫サイトカイン・脳シグナル伝達機構を終板器官・視床下部系で明かにする。(2)(1)で変換された血中サイトカイン由来脳信号が視床下部のどこで、何を媒介物質としてどのように情報処理され自律神経系に伝達されるかという視床下部内機構を解明し、(3)その自律神経を介する免疫系制御機構を明かにすること、さらに(4)視床下部におけるpituitary adnylate cyclase activating polypeptide(PACAP)の神経内分泌系への作用を検討することである。研究成果:以下のような成果を挙げることができた。また、その成果を第1回ストレス学会において報告討議した。1.麻酔下ラットの第3脳室壁前腹側領域(AV3V)ニューロンの活動が、血中に投与したIL-1βに反応して発火頻度を変化させること、およびそのニューロンが視索前野(POA)た室傍核(PVN)へ情報を送っていることを明かにした。さらに、血中IL-1βによるニューロン活動の変化が、局所でのプロスタグランディンE2(PGE2)の産生を介していることを示した。2.脾臓における細胞性免疫の制御に関与する脾臓交感神経の活動が、脳室内CRFおよびPGE2の投与によって著明に上昇することを明かにした。PGE2受容体のサブタイプであるEP1、EP2、およびEP3受容体のうち、それぞれ特異的なアゴニストの投与に対する脾臓交感神経活動の変化を検討し、PGE2の作用はEP1受容体を介していることが明かになった。3.ラットに水浸拘束ストレスを加えると、血中IL-6が増加することを明かにし、その機序として肝臓におけるCRF系が重要であることを示した。4.視床下部PVNの大細胞領域ニューロンの活動がPACAPによって長時間にわたった上昇し、その効果は、VIPアンタゴニストでブロックされずPACAPに特異的な受容体を介したものであることを示した。
