Research Abstract

細胞が、がん化して浸潤能や転移能を獲得する過程において、細胞の形態や運動性、極性、接着性、分泌性等々の異常が重要な役割を果たしているが、これらの細胞機能は主としてアクチン系の細胞骨格により制御されている。一方、Rho低分子量G蛋白質が、私共を含めたいくつかのグループによって、アクチン細胞骨格の再編成を介して、上述した細胞機能を制御していることが明らかにされつつあり、Rhoを介するシグナル伝達経路の異常が、がん細胞の浸潤や転移において重要な役割を果たしている可能性が高い。本研究では、Rhoの活性化機構と作用機構に焦点をしぼって解析を行い、以下の成果を得た。私共は、アクチン繊維を細胞膜へAnchoringするERM(Ezrin,Radixin,Moesin)-CD44系が、Rhoの作用点として機能していることを明らかにしているが、このERMが、Rho-Rho GDI複合体に直接作用し、RhoをRho GDIから解離させて活性化反応を開始させる作用も有していることを明らかにした。一方、活性化されたRhoは、BNI1様標的蛋白質に作用し、アクチン結合蛋白質であるプロフィリンを介してアクチンの重合過程を制御すると共に、何らかの標的蛋白質を介して、ERMの細胞膜への移行過程、すなわちCD44との会合を制御することにより、アクチン細胞骨格の再編成を統合的に制御していることを明らかにした。以上の成果を得たことにより、本年度の研究は順調に進んだと考えている。
