Research Abstract |
Unicellular algae, Cyanidium caldarium and Galdieria sulphuraria were isolated from a acid-hot springs, Nasu, Yumoto spa, Tochigi prefecture in 1994. Blue-green algae (Cyanobacteria), Synechococcus, Phormidium and Oscillatoria were found in hot-springs at neutral pH, such as Atagawa, Mine and Shuzenji spas, Izu peninsula in 1995. Then, total DNA were extracted from C.caldarium, G.sulphuraria and C.merolae and 18s rRNA genes of them were multiplied by using PCR.As a result, the gene was much obtained from C.caldarium. Partial DNA sequence of the gene of C.caldarium are considerably different from those of other red algae. We postulate that C.caldarium may be far from standard red algae phylogenetically.
