Research Abstract

In vivoおよびIn vitroの研究で乳癌の発生、増殖、転移に与えるリノール酸、EPAおよびDHAの影響を検討し、次の結果を得た。
(1)リノール酸は、マウスに移植したホルモン非依存性乳癌(MM48)の増殖および転移を促進し、EPAおよびDHAはそれらを抑制した(Kinoshita andNoguchi: Inhibitory effects of purified eicosapentaenoic acid and docosahexaenoic acid on growth and metastasis of murine transplantable mammary tumor. (2)リノール酸は、In vitroでヒト乳癌細胞(MDA-MB-231)のProstaglandin EやLeukotriene Bの分泌、および細胞増殖を促進した。しかし、EPAおよびDHAはそれらを抑制し、EPAの抑制はn-3/n-6比が1:0.69、DHAのそれはn-3/n-6比が1:2.08以上で認められ、更にそれらの抑制は培地中のLeukotriene BよりもProstaglandin Eの濃度と相関することが明かとなった(Noguchi et al: Effects of eicosapentaenoic acid and docosahexaenoic acid on cell growth and prostaglandin E and leukotriene B production by a human breast cancer cell line(MDA-MB-231). (3)リノール酸は、In vitroでヒト乳癌細胞(MDA-MB-231およびMCF-7)の増殖を促進し、更にMCF-7乳癌細胞ではc-mycの発現を促進することが明かとなった(Earashi, Noguchi et al.: Effects of linoleic acid and eicosanoid synthesis inhibitors on the growth and c-myc oncogene expression of human breast cancer cell. Int J Oncol 8:145-151,1996)。
