Research Abstract

LH/hCGレセプターは、スプライシングの相違により最低二種類の分子量を持つレセプターが発現されている可能性がありこれらの相違が結合能、情報伝達系にどのような影響を与えるか検討した。この結果LH/hCGレセプターは、スプライシングの相違により生じると思われる短い型のレセプターを発現させたCHO cellではcAMPの上昇が見られなかった。このことは、レセプター合成のdown regulationとして機能している現象とも考えられた。
一方、生理的条件下では両方のレセプターが同時に発現されている可能性があるので、COS細胞に2つのレセプターを同時に発現させたものと、wild typeのみの発現実験との比較を試みた。現在までのところ、短いレセプターの発現が長いものに対して抑制的に働くことが分かった。このことはスプライシングの相違により、レセプター機能の調節が行われている可能性を示すものである。
