Research Abstract

1)サルの系:エイズ脳炎の制御を目的として、血液脳関門を通過する6-cl-ddGを、SIVmac239接種により神経系の異常が見られるサルに投与したところ、症状の回復とウイルス量の低下がみられた(向井)。SIVmac239の経膣感染を行ったところ、感染初期にPBMCで検出されるウイルスは一部のポピュレーションに限局されており、また多様性のあるウイルスに感染しているサルの膣内で検出されるウイルスも限局されていた。このことは膣においてバリアーがあることが明らかになった(速水)。invivo継続SHIVはサルPBMCでの増殖能が強くなり、その接種ザルでエイズ様死がみられ、リンパ節病変にウイルスゲノムが検出された(速水)。
2)マウスの系:hu-PBL-scidにおけるHIVの感染性は、ウイルスの細胞親和性により異なり、マクロファージ親和性HIV-1株がT細胞株親和性HIV-1よりも強い増殖性と病原性をもつことが判った(田中)。TNF-α欠損HIV遺伝子導入マウスにLPSを投与したところ、HIV遺伝子活性化の抑制が見られ、TNF-αがLPSによるHIV遺伝子活性化の主要なメディエターであることが解った(岩倉)。CD4とコレセプターCXCR4を共発現するトランスジェニックマウスを作製し、HIV感受性を調べたところ、プロウイルス化の段階までは感染を支持することが明らかになった(岩倉)。ブルトン型チロシンキナーゼ(Bt1)遺伝子のMAIDS発症における役割を明らかにする目的で、変異Btkトランスジェニックマウス(Tg)を作製したところ、XIDマウスに類似したB細胞異常を示し、MAIDSウイルス感染による脾腫、高ガンマグロブリン血症等のMAIDA徴候の発現の遅延が認められた。
3)ネコの系:FIVのAP-1結合部位およびvif,orf-A遺伝子の変異体をネコに接種し、AP-1結合部位と各遺伝子の役割を解析したところ、各変異体は、AP-1、orf-A、vif変異体の順に生体内でのウイルス増殖の遅延が見られた。FIV接種ネコにおいて末梢血中では通常少ないCD8α陽性細胞が増加していた(見上)。
