Research Abstract

Candida albicans is a medically inportant dimorphic yeast which causes human mycotic infections. The components of the cell wall of C.albicans are beta-glucan, chitin and mannan, although some aspects of their biosynthesis have been studied, the molecular organization of the cell wall is largely unknown. Using the regenerating protoplast of C.albicans, occurrence and deposition of beta, 1,3-glucan, chitin and mannan in the wall were investigated at the electron microscope level by immuno-glod and wheat germ agglutinin (WGA) -gold. We demonstrated that fibrillar structures associated with specific binding of immuno-gold and WGA-gold were formed over the cell surface at an early stage of regeneration, suggesting that newly synthesized fibrillar structures were composed of both beta-1,3-glucan and chitin. Reactivity to anti-C.albicans mannan was detected primarily on the surface of cytoplasmic membrane of freshly prepared protoplasts. At tshe middle stage of regeneration, mannan was embedded within and over the fibrillar network of beta-glucan and chitin, and finally localized on the surface of the completed cell wall. These data suggest that mannan molecules are produced in the cytoplasmic membrane and then are transferred through a network of microfibrils to the surface of the cell wall.
